Promising Early Results Reported for Russia’s Pioneer mRNA Melanoma Vaccine

Russian medical scientists have reported encouraging early clinical data from the trial of a locally developed mRNA-based cancer vaccine targeting advanced melanoma. According to researchers, the primary patient undergoing the novel treatment has developed a targeted immune response capable of combating malignant cells without experiencing severe adverse side effects.

The bespoke therapeutic vaccine, named Neooncovac, was created through a joint effort involving the Gamaleya Research Institute of Epidemiology and Microbiology—the institution behind the Sputnik V COVID-19 vaccine—alongside the National Medical Research Centre of Radiology and the Blokhin National Medical Research Centre of Oncology. Aleksandr Gintsburg, scientific director of the Gamaleya Institute, verified that the patient’s immune system is generating biological compounds specifically designed to assist white blood cells in detecting and destroying primary tumours as well as metastatic growths.

The trial involves a 60-year-old male patient from the Kursk region diagnosed with stage-three cutaneous melanoma, currently receiving treatment at the Herzen Moscow Oncology Research Institute. Neooncovac was first administered to the patient in April, marking the initial deployment of Russia’s newly approved personalized mRNA cancer therapies. The therapeutic regimen consists of a total course of up to ten intramuscular injections, with additional doses planned while clinicians continuously evaluate the patient’s long-term immunological profile.

Concurrently, Russian laboratories are preparing custom mRNA formulations for ten additional candidates, with individual manufacturing cycles taking between six and twelve weeks. Detailing the mechanism behind the therapy, Andrey Kaprin, head of the National Medical Research Centre of Radiology, explained that each dose is formulated following genetic sequencing of both the patient’s tumor and healthy tissue. This bioinformatic mapping identifies specific neoantigens, effectively training the host’s immune system to neutralize cancerous cells while sparing healthy tissue.

Late last year, the Russian Ministry of Health granted authorization for two distinct personalized cancer treatments: Neooncovac for advanced skin melanoma and Oncopept, a peptide-based vaccine designed for metastatic colorectal cancer. Health authorities confirmed that patients receiving Oncopept have similarly demonstrated positive immune activation. Building upon these initial clinical milestones, research teams are expanding the mRNA platform to develop targeted immunotherapies for pancreatic, renal, non-small cell lung cancers, and glioblastoma.
